Laboratory of confocal microscopy
The Microscopy Laboratory at the Department of Botany, Faculty of Science UPOL was opened on December 19, 2006 with the support of the research project (Ministry of Education, Youth and Sports of the Czech Republic) MSM 6198959215 "Variability of components and interactions in plant pathosystems and the influence of environmental factors on their manifestation" as a reference workplace of the company Olympus Czech Group, s.r.o.
Equipment
Laser scanning confocal microscope Fluoview 1000
System includes:
Inverted system microscope Olympus IX 81
- Observation in transmitted light, Nomarsky DIC - condenser with long working distance
- Fluorescence - excitation with 100W Hg discharge lamp, fluorescent filters (WU, WG, NB, LSM)
- Motorized focusing and lens exchange system (10, 20, 40, 60, 100x)
Olympus FV 1000 imaging system
- Laser unit with 4 lasers with wavelengths in the range UV–RED (405–633nm) and AOTF laser combiner
- Light sources:
- Red (He–Ne 633nm / 10mW),
- Green (He–Ne 543nm / 1,5mW),
- Blue (Ar 457nm, 488nm, 515nm / 30mW),
- near UV (diode pumped, 405nm / 20mW)
- 3 detection channels for confocal fluorescence image detection + transmitted light detector/Nomarski DIC

Funding rules
Use of the microscope by staff of the Faculty of Science departments participating in its purchase (KB, KBBG, KBC, KEF)
The operation of the confocal microscope will be financed in advance by the Department of Botany, Faculty of Science, University of Olomouc, which will then re-account the costs, reduced by funds received from fees from other institutions (see below), every 3 months to the departments that participated in the purchase of the microscope (i.e. KB, KBBG, KBC, KEF), in proportion to the use of the device by the individual departments during the given period.
In the case of expensive repairs (e.g. overloading of lasers, etc.), the participating departments will agree on the method of their payment in proportion to the use of the device during the period since the last repair (replacement) of the relevant part of the microscope.
Fees for the use of the microscope by staff of other departments within the Faculty of Science and Faculty of Medicine UPOL and other institutions
In case of free capacity, the microscope will also be available to employees of other UP departments and other institutions, provided that they pay fees for the operating hours used. Departement of Botany will invoice users for fees in the amount corresponding to the number of operating hours used, with each half hour started being counted.
The rates will be different for UPOL employees and for employees of other workplaces:
- Departments of UPOL (except KB, KBBG, KBC, KEF): 400 CZK/hour
- Institutions outside UPOL (in this case, energy costs and the technician's salary will also be included in the costs): 600 CZK/hour
The above rates may be subject to change after approval by representatives of the Faculty of Science departments participating in the purchase of confocal microscope (KB, KBBG, KBC, KEF).

Laboratory of flow cytometry
The laboratory is fully equipped for analyzing the absolute amount of DNA in the nucleus (genome size) and ploidy level, determining the ratio of GC:AT bases in the genome, and determining the method of seed formation (sexual or asexual).
